Verteporfin-PDT treatment induces punctate regions of cytotoxicity concomitant with architectural breakdown of a 3D tumor micronodule. Combined with high content image analysis, this in vitro tumor platform reports a quantitative shift in size distribution towards smaller nodules (Celli et al. J Biomed Opt 2010), suggestive of an increased sensitivity to chemotherapy. Leveraging this 3D system to screen combinations of PDT with traditional chemotherapy reveals a sequence-dependent synergistic enhancement of carboplatin efficacy in ovarian micronodules (Rizvi et al. Cancer Res 2010).
